DNA-Interactive Drugs: Neocarzinostatin

Name Neocarzinostatin
Zinostatin
CAS#  9014-02-2
NCS#  69856
MW Apoprotein 10,700 (tightly associated non-protein chromophore MW ~659) [Meienhofer, 1972; Ishida, 1965].
Category Enediyne-containing peptide antibiotic
Binding Mode Intercalation of napthoate moiety [Povirk, 1981]
Site Specificity Adducted bases occur T>A>>C>G [Poon, 1977; Golik, 1987], oligopyrimidine runs cleaved preferentially [Sugiura, 1989].  UV-activated ncs has increased activity against G [Uesawa, 1989].
Effect on DNA Strand scission agent, activated by reducing agents or UV irradiation.  Intercalation unwinds DNA by 21o, each molecule lengthens DNA by 0.33 nm [Povirk, 1981].
Source Originally extracted from Streptomyces carzinostaticus var. F-41 [Ishida, 1965].
Preparation and Use 200 microM stock prepared in ddH2O
