| Category |
Anthracycline |
| Binding Mode |
Threading intercalation; glucosamine in major groove, anthracycline
core intercalated, nogalose sugar and methyl ester in minor groove [Arora,
1983; Williams, 1990]. |
| Site Specificity |
Regions of alternating Y-R sequence: 5'-(C>T)(G>A)-3' based on DNase
I footprinting [Fox, 1986; Fox, 1988]. X-ray crystal studies show
nogalamycin intercalated between 5-CpG-3' [Williams, 1990; Liaw, 1989;
Gao, 1990]; NMR solution structure shows nogalamycin intercalated between
5'-TpG-3' [Searle, 1988], 5'-CpA-3' [Zhang, 1990], and 5-CpG-3' [Robinson,
1990 ; van Houte, 1993]. Nogalamycin appears not to bind DNA at 4oC
[Fox, 1986]. |
| Effect on DNA |
Unwinding angle of 18o [Waring, 1981]. May unbend
intrinsically bent DNA [Cons, 1990]. Base pairs surrounding drug
are strongly distorted [Williams, 1990]. Association with DNA likely
to require local melting [Collier, 1984] |
| Source |
Originally isolated from Streptomyces nogalator var. nogalator
[Bhuyan, 1965] |
| Preparation and Use |
Prepare in DMSO using extinction 1.12x104 M-1
cm-1 at 475 nm [Das, 1974]. Drug is light sensitive.
Sparingly soluble in alcohol or water. |
