DNA-Interactive Drugs: Amiloride

Name Amiloride HCl
Midamor
3,5-diamino-6-chloro-N-(diaminomethylene) pyrazinecarboxamide
MW 229.65
Category Pyrazine derivative
Binding Mode Intercalation.
Site Specificity A/T preference, by DNA cleavage inhibition [Bailly, 1993].
Preparation and Use E1%1cm at 285 nm is 555 OD, and E1%1cm at 362 nm is 617 OD (ie, 1g of this compound in 100 ml total volume would have an absorbance of 555 O.D. units at a wavelength of 285 nm, measured with a 1 cm light path).    Tablets have been found not to dissolve directly in DMSO.  Drug should be dissolved in the minimal amount of ddH2O necessary, and then brought to a final volume in DMSO.  The solution can then be filtered, first through Whatman paper, then through a nitrocellulose filter attached to a syringe.  The resulting solution is yellow.
